Abstract

Coffee cherry waste is a type of agroindustrial residue that contains lignocellulose, making it relatively difficult to decompose naturally. The exploration of lignocellulose-degrading microorganisms is a potential approach to supporting sustainable agricultural waste management. This study aimed to isolate and conduct preliminary screening of microorganisms from coffee fruit peel waste based on qualitative cellulolytic and ligninolytic activities. Microorganism isolation was performed using the serial dilution method and colony purification. Cellulolytic activity was observed based on the formation of a transparent zone on carboxymethylcellulose (CMC) medium, while ligninolytic activity was observed based on a color change on tannic acid medium. The study yielded 29 microbial isolates, consisting of 14 fungal isolates and 15 bacterial isolates. Based on colony morphology, the fungal isolates were tentatively classified into the genera Aspergillus sp., Penicillium sp., and Trichoderma sp., while the bacterial isolates were tentatively classified into the genera Bacillus sp., Azotobacter sp., and Pseudomonas sp. Several groups of isolates showed a positive response to screening for cellulolytic and ligninolytic activity based on visual indicators in the test media. The results of the study indicate that coffee fruit peel waste has the potential to serve as a source of lignocellulolytic microorganisms that can be further investigated through molecular characterization and quantitative enzyme activity analysis.
